Principles of Biology : Laboratory
Lab 14 : DNA
Theme: Look at the theme I gave
you in Lab 7 and write a similar, short, concise theme for our protein
fingerprinting lab.
Materials and Methods:
Attached: "Extremely Easy DNA Extraction" - from
Carolina Biological. And Lab 6B in the "Biology Lab Manual",by The
College Board, 2001. <cite correct page
numbers>
Results: The data is presented
online at http://www.siskiyous.edu/class/bio1/lab14/
Note: You have not been provided with the known values of EcoRI and so
you can skip those questions relating to how close your approximations
came. Also, notice the stirkout in the next paragraph. I decided not to
include those questions, but forgot to remove these instructions from
this page. :(
Analysis and Conclusion Section : Answer the questions and
construct and use the graph that are
included in the lab book for part B of Lab 6. Answer the questions in
the Easy DNA Extraction handout. Also, include
the following answers in a paragraph concerning other applications of
the
technique:
1) Why is untreated ("genomic" or plasmid) DNA unsuitable for DNA
fingerprinting? What is the treatment, say of plasmid DNA, to get it
ready for electrophoresis?
2) How is DNA fingerprinting achieved for human DNA? (see text book).
